SUMMARY:Integration Site Analysis During Long Term Follow-Up for Gene Therapies with Integrating Viral Vectors
DESCRIPTION:The FDA’s Center for Biologics Evaluation and Research (CBER) Office of Therapeutic Products (OTP) is hosting a virtual scientific public workshop on the clinical use of integration site analysis (ISA) during long term follow-up following administration of gene therapies with integrating viral vectors. \nIn this workshop\, FDA will convene a panel of external experts to discuss the risk of insertional mutagenesis and best practices for ISA method design\, data analysis\, and clinical interpretation. The workshop participants will include research scientists with experience in vector design and ISA\, as well as clinician-scientists with expertise in clinical interpretation of ISA data. \nFocus for this Workshop\nAs of 2024\, twelve approved and hundreds of investigational cell-based gene therapies use integrating viral vectors. While stable integration may have considerable therapeutic benefits\, integration into genomic DNA can alter host gene expression and may contribute to secondary malignancies via insertional mutagenesis. Additionally\, the choice of vector (e.g.\, lentivirus versus gammaretrovirus) and cell substrate (e.g.\, T cells versus CD34+ cells) may also contribute to this risk. \nISA is the established approach to identify vector insertion sites. However\, ISA method design and data interpretation are highly complex and product-specific. Moreover\, ISA methods may be costly and challenging to perform. SUMMARY:G20 Summit Rio 2024
DESCRIPTION:The G20 Leaders’ Summit is scheduled for 18-19 November 2024\, in Rio de Janeiro\, with the presence of the leaders of the 19 member countries\, plus the African Union and the European Union. \nThe G20 brings together the countries with the largest economies in the world. The member states meet annually to discuss economic\, political and social initiatives. The group defines itself as the main forum for international economic cooperation. \nThe G20 countries account for half the world’s land mass\, two-thirds of the world’s total population\, and a staggering 85% or more of global GDP. These imply both a large magnitude of health burden on the one hand and a depth of experience and strong base of resources on the other. The G20 Health Working Group was established under the German Presidency in 2017 to develop a shared international agenda on issues such as strengthening health-care systems\, reducing malnutrition\, health-crisis management\, and scaling up the fight against pandemics; these are India’s domestic priorities too. \nIn addition to the summit\, more than 100 meetings of working groups and task forces will comprise the G20 event\, both face-to-face and virtual\, at technical and ministerial level\, in the host cities of Brasil’s five regions. The highlight will be the Summit in Rio de Janeiro. \nThe participating countries are Argentina\, Australia\, Brasil\, Canada\, China\, France\, Germany\, India\, Indonesia\, Italy\, Japan\, Mexico\, Republic of Korea\, Republic of South Africa\, Russia\, Saudi Arabia\, Turkey\, United Kingdom and the United States of America\, African Union and European Union. In addition\, invited guest countries and international organizations participate in the G20. Media reports indicate the Presidents of Russia\, China and Mexico will not be in attendance. State leaders from the other member states will likely attend. \nThe G20 troika system was officially adopted at the Cannes Summit (France) in 2011 as the framework for steering the G20 process. The “troika” consists of the current\, previous\, and next G20 presidencies and the three members cooperate with one another in preparing for the G20 Summit. SUMMARY:What Research is Important to Prepare for H5N1 Outbreaks?
DESCRIPTION:The World Health Organization will host a virtual meeting on 19 March to identify the knowledge gaps and priority research questions related to the development and evaluation of medical countermeasures to prepare for and respond to H5N1 outbreaks \nThe meeting will outline regulatory pathways for mRNA vaccines and other novel approaches and review other therapeutics\, including monoclonal antibodies and long-acting antivirals. \nH5N1 influenza virus infection can cause a range of diseases in humans\, from mild to severe and in some cases\, it can even be fatal. Symptoms reported have primarily been respiratory\, but conjunctivitis and other non-respiratory symptoms have also been reported. There have also been a few detections of A(H5N1) virus in persons who were exposed to infected animals or their environments but who did not show any symptoms. \nThe goose/Guangdong-lineage of H5N1 avian influenza viruses first emerged in 1996 and has been causing outbreaks in birds since then. Since 2020\, a variant of these viruses has led to an unprecedented number of deaths in wild birds and poultry in many countries. First affecting Africa\, Asia and Europe\, in 2021\, the virus spread to North America\, and in 2022\, to Central and South America. From 2021 to 2022\, Europe and North America observed their largest and most extended epidemic of avian influenza with unusual persistence of the virus in wild bird populations. \nSince 2022\, there have been increasing reports of deadly outbreaks among mammals also caused by influenza A(H5) – including influenza A(H5N1) – viruses. There are likely to be more outbreaks that have not been detected or reported. Both land and sea mammals have been affected\, including outbreaks in farmed fur animals\, seals\, sea lions\, and detections in other wild and domestic animals such as foxes\, bears\, otters\, raccoons\, cats\, dogs\, cows\, goats and others. \nWHO works with countries to help them prevent\, detect and manage cases of H5N1 virus infection in humans when they occur. WHO collaborates with global health partners and agencies\, including the World Organization for Animal Health (WOAH)\, and the Food and Agriculture Organization of the United Nations (FAO)\, to monitor the risk\, and control and prevent the spread of animal diseases. \nWHO’s global laboratory system\, the Global Influenza Surveillance and Response System (GISRS)\, identifies and monitors strains of circulating influenza viruses\, and provides advice to countries on their risk to human health and available treatment or control measures. WHO works with partners around the world on pandemic preparedness for H5N1 and other animal influenzas. \nInfluenza H5 pathogens have been included in the list of priority pathogens by the WHO R&D Blueprint for Epidemics. To coordinate efforts for research preparedness of H5N1 outbreaks\, this global consultation will review current and novel vaccines and preventive pharmaceuticals. \nPlease visit the World Health Organization’s website for more information and to register.

DESCRIPTION:As part of Advancing Drug Discovery: A Webinar Series of the National Academies of Sciences\, Engineering and Medicine\, a session on Unique Capabilities of Cryo-EM in Structure Determination and its Potential in Drug Discovery will be held on March 25th\, 2025. \nCryo-EM has emerged as a powerful structural technique that is generally applicable to samples spanning from single proteins to large viruses\, including integral membrane proteins and large macromolecular assemblies\, even when they are flexible or involve transient\, regulatory interactions. The minimal requirement concerning sample amount and the capacity to describe conformational ensembles make cryo-EM the technique of choice when characterizing samples that challenge methods like NMR or crystallography. \nThe resolution of the structures obtained by cryo-EM has been increasing with improvement in hardware and software\, making it relevant in the drug discovery arena. \nDr. Eva Nogales\, Distinguished Professor in Molecular and Cell Biology at the University of California\, Berkeley\, will introduce the technique and demonstrate its capability in studies of the microtubule cytoskeleton\, molecular machinery involved in gene expression regulation\, and other biological systems that had defied structural characterization by more traditional methods. \nPlease visit the National Academies’ Unique Capabilities of Cryo-EM in Structure Determination and its Potential in Drug Discovery event page for more information and to register.

SUMMARY:Building Trust in the H5N1 Response: Perspectives from the Field
DESCRIPTION:The National Academies will host a event on Building Trust in the H5N1 Response: Perspectives from the Field via webcast on March 27\, 2025. \nAvian influenza is a growing threat to both human and animal health. In March 2024\, avian influenza (H5N1) was detected for the first time ever in dairy cattle. The ongoing H5N1 outbreak in the United States has impacted livestock\, diminished livestock production\, decimated wildlife\, and resulted in human infections. The potential for viral mutation and transmission poses a continued threat to human health. Despite the urgency of this evolving situation\, response has been hindered by a lack of trust among key stakeholders. Varying policies at national\, state\, and local levels have led to challenges in mitigation efforts. Following a National Academies workshop on potential research priorities to inform the H5N1 response\, the National Academies Forum on Microbial Threats will host a webinar in which agriculture producers and workforce health specialists will discuss their perspectives on steps that would build greater trust between producers and health officials to enable a more coordinated One Health response. \nThe Forum invites individuals who have been involved in response to the ongoing North American H5N1 outbreak to share their experiences and perspectives on building trust between agriculture producers and public health officials. Comments may be used to develop discussion questions and identify key topics of discussion for the webinar. DESCRIPTION:The National Academies will host a public briefing Assessing and Navigating Biosecurity Concerns and Benefits of Artificial Intelligence Use in the Life Sciences on April 2\, 2025. \nThe Committee on Assessing and Navigating Biosecurity Concerns and Benefits of Artificial Intelligence Use in the Life Sciences will brief on the report\, The Age of AI in the Life Sciences: Benefits and Biosecurity Considerations. \n\n\n\nArtificial intelligence (AI) applications in the life sciences have the potential to enable advances in biological discovery and design at a faster pace and efficiency than is possible with classical experimental approaches alone. At the same time\, AI-enabled biological tools developed for beneficial applications could potentially be misused for harmful purposes. Although the creation of biological weapons is not a new concept or risk\, the potential for AI-enabled biological tools to affect this risk has raised concerns during the past decade. \nThis report\, as requested by the Department of Defense\, assesses how AI-enabled biological tools could uniquely impact biosecurity risk\, and how advancements in such tools could also be used to mitigate these risks.
